李悦,博士。2011-2014美国Nebraska病毒学中心完成博士后研究,美国NIH Fogarty项目长期受训人员,南开大学艾滋病研究中心(CFAR-NK)秘书。主要从事艾滋病及相关病毒的感染性动物模型建立以及艾滋疫苗的研发和保护效果评价等相关研究,工作重点为HIV/AIDS人源化小鼠动物模型的建立及免疫病理研究,HIV/SIV嵌合病毒的构建及中国源恒河猴模型的建立,复制及非复制型HIV疫苗保护效果的评价及免疫效应机理研究。参加国家科技重大专项、973课题、科技部国际合作重大项目、自然科学基金等项目。在Angewandte Chemie, PNAS, Journal of Leukocyte Biology, Virology, Virus Research, Journal of Virological Methods, Virus genes和Virologica Sinica等期刊上发表多篇学术论文。代表性研究成果有:成功构建一株可利用非天然氨基酸编码调控的HIV-1减毒活疫苗,对新型艾滋病疫苗的研发具有重要意义;建立并完善了多个中国HIV-1流行亚型CRF-07BC SHIV/恒河猴动物模型,并在此基础上针对中国复制型天花病毒载体艾滋病疫苗的保护效果进行临床前评价,同时也对宿主产生的免疫反应以及病毒超感染的机理进行了探讨。证明rTV单独免疫产生的保护效果较DNA初免rTV加强的免疫策略更好,同时验证了感染病毒的毒力和亚型对HIV-1超感染能否保护具有重要影响;参与建立了以NSG为基础的新型BLT人源化小鼠,该模型为国际上公认的最接近人体免疫系统的新一代人源化小鼠模型。
With more than 10 years of work experiences HIV-1/AIDS, including vaccine development and effect detection, AIDS animal model generation, virus and host immune responses. From 2011 to 2014, my research focused on a novel HIV-1 live-attenuated vaccine construction through genetic code expansion, which will be tested in BLT-humanized mouse, a HIV-1 animal model I involved to generate. From 2004 to 2010, based on replication-competent Tiantan vaccinia, I determined HIV-1 vaccines which could protect Chinese rhesus macaques from simian HIV infection. Meanwhile, I established the SHIV/macaque model to test candidate AIDS vaccines targeting HIV-1 strain, combined replication-competent SHIVs version containing the Chinese HIV-1 env gene and identification of infectious activity. Moreover, to figure out host immune response after HIV-1 infection, I sought to study potential mechanisms after mucosal infection or antiviral therapy of HIV-1 by SIV/macaque and humanized mouse model, such as immune cells characterization and how antibodies to protect. By now, as an Associate Professor in Department of Microbiology in the College of Life Sciences, Nankai University, I am mainly working on the HIV-1/AIDS antibody-drug conjugate development and host immune responses during infection.
